The 5h4 antibody monoclonal antibody specifically binds to mouse cd122. Cd122 is a 70-85 kda type i transmembrane glycoprotein of the hematopoietin receptor superfamily. Cd122 is also known as il-2 receptor beta chain (il-2r β ) or il-15 receptor beta chain (il-15r β ) as it helps form signaling receptor complexes for interleukin-2 (il-2) or il-15, respectively. Cd122 can combine with either cd132 (γc) alone or cd132 plus cd25 (il-2rα) to form inte rmediate or high-affinity il-2 receptor complexes, respectively. The il-2r β chain is constitutively expressed on nk cells and at lower levels on some thymocytes, t cells, b cells, monocytes, and macrophages. Il-2r β chain expression is up regulated by il-2. The immunogen used to generate this hybridoma was rat myel oma cells transfected with a truncated il-2r β cdna. The antibody was conjugated to bd horizon bv421 which is part of the bd horizon brilliant™ violet family of dyes. With an ex max of 407-nm and em max at 421-nm, bd horizon bv421 can be excited by the violet laser and detected in the standard pacific blue™ filter set (eg, 450/50-nm filter). Bd horizon bv421 conjugates are very bright, often exhibiting a 10 fold improvement in brightness compared to pacific blue conjugates.
